The show was filmed at Camp Pendleton, Desilu Studios's Desilu-Cahuenga, and RKO Forty Acres backlot, where The Andy Griffith Show was filmed. Though Ruben preferred the use of a multiple-camera setup for comedy programs, Gomer Pyle used a single-camera setup because much of the shooting was conducted outdoors. In his book And The Show Goes On, Sheldon Leonard explained that the armed forces offer levels of "cooperation" with filmmakers. Because the Marines felt that the show would be good for the branch's image, Gomer Pyle was given "total cooperation," meaning that the show was allowed unlimited access to military equipment.

The cousin of Gomer Pyle, Goober was initially often referred to by Gomer but never seen until his appearance in the episode "Fun Girls," which was the only Andy Griffith Show episode in which the two appeared together. Goober later appeared in an episode of Gomer Pyle, USMC.

Gomer Pyle is a fictional character played by Jim Nabors and introduced in the middle of the third season of The Andy Griffith Show, A naïve and gentle auto mechanic, he became a recurring character in the January 1963 episode "Man in a Hurry"   Nabors played Pyle for 23 episodes, from 1962 to 1964. After two seasons on The Andy Griffith Show, the character was spun off to Gomer Pyle, U.S.M.C. in 1964 which ran until 1969.

When he saw the episode of The Danny Thomas Show featuring Andy Taylor, he called Griffith suggesting that his sheriff character might reasonably need a deputy. Griffith liked the idea and suggested that he call Executive Producer Sheldon Leonard. Griffith later recalled that Don Knotts' contribution was the show's saving grace because he was uncomfortable with the original concept to have Andy Taylor being the comic lead. In an interview with The Archive of American Television, Griffith admitted,
