The unit reactivated on 1 June 1940 at Fort McClellan, Alabama, before inactivating on 1 March 1946 at Camp Butner, North Carolina. It reactivated 15 July 1947 at Fort Ord, California. It reorganized and was redesignated on 1 April 1957 as Headquarters and Headquarters Company, 1st Battle Group, 22nd Infantry and remained assigned to the 4th Infantry Division (with its organic elements being concurrently constituted and activated). It was reorganized and redesignated on 1 October 1963 as the 1st Battalion, 22nd Infantry. It was deployed to Vietnam as part of the 2d Brigade of the 4th Infantry Division and conducted operations in the Central Highlands.

As part of the 1st Cavalry Division, 2-8th Cavalry was sent to Vietnam in September 1965. The Mustangs saw some of the fiercest battles. B Company was the first unit in the 1st Cavalry Division to receive the Valorous Unit Citation in 1966. 2-8th Cavalry actively patrolled the jungles of Vietnam to the north and south of Highway 1, east of Bien Hoa, until it stood down in June 1972. On 10 May 1972, a CH-47 Chinook helicopter carrying a platoon from D Company, 2-8th Cavalry (named Angry Skipper) crashed and all aboard were killed. The Battalion left Vietnam in 1971 and was inactivated on 28 June 1972.

In August 1965, elements of the 35th Infantry started their rotation into the Vietnam War, arriving in South Vietnam. The 1st and 2d battalions, along with the 1st Battalion, 14th Infantry, and the 2d Battalion, 9th Artillery provided the core combat units of the 3d Brigade, 25th Infantry Division and were deployed to the Central Highlands at Pleiku. 1st Battalion, 14th Infantry: In the fall of 1965 the 25th Division was ordered to Vietnam. The initial brigade to go was the 3rd Brigade composed of the 1st Battalion, 14th Infantry, the 1st Battalion, 35th Infantry, the 2nd Battalion, 35th Infantry and the 2nd Battalion, 9th Artillery. The urgency of getting the 3rd Brigade to the Central Highlands of Vietnam led to the Army and Air Force undertaking Operation Blue Light, a massive airlift of the entire 3rd Brigade from Hawaii to Pleiku. The airlift began on 28 December 1965 and was successfully concluded on 17 January 1966.

Three battalions (2nd Bn, 19th Arty; 1st Bn, 21st Arty; and 1st Bn, 77th Arty) were 105mm towed battalions armed with the new M102 howitzer. The 2nd Bn, 20th Arty was an Aerial Rocket Artillery (ARA) battalion, consisting of three batteries, each with twelve UH-1B helicopters firing 2.75-inch rockets, while Btry E, 82nd Arty was an aviation battery with 20 helicopters for observation and other aviation support. The 2nd Battalion of the 11th Artillery Regiment ("Dragon Artillery") was a towed 155mm Howitzer Battalion at Dau Tieng which saw service first as part of II Field Force Vietnam Artillery and later with Task Force Oregon. It was deploued to Duc Pho on 19 April 1967, Bien Hoa in January 1968 and on 8 March 1968 to I CTZ and 23rd Artillery Group's control at Gia Le. On 10 June 1968 it was assigned to 101st Airborne Division.
